GlycanPac Series

The Thermo Scientific™ GlycanPac™ AXR-1 and GlycanPac AXH-1 columns are high-performance, silica-based HPLC columns designed for high-resolution glycan separations. They provide industry-leading resolution with unique selectivities for biologically important glycans, either labeled or native, using fluorescence and/or mass spectrometry (MS) detection.  Both columns separate glycans by charge and size. Additionally the GlycanPac AXR-1 columns are superior for separating glycan isomers. The GlycanPac AXH-1 columns are also used for fast, accurate, and simple quantification of glycans by charge.


The GlycanPac AXR-1 column is based on innovative mixed-mode surface chemistry that combines both weak anion-exchange (WAX) and reversed-phase interaction mechanisms to produce exceptional glycan selectivity and industry-leading resolution. The WAX functionality provides retention of negatively charged glycans that elute in order of increasing charge, and the reversed-phase interactions facilitate high-resolution separation of glycans of the same charge according to their isomeric structure and size. These attributes produce unparalleled glycan separations. The GlycanPac AXR-1 column is designed and tested for HPLC and UHPLC methods using either fluorescence or MS detection applications, and uses volatile aqueous buffers (e.g., ammonium acetate or ammonium formate) and acetonitrile, presenting the eluting glycans ready for introduction into MS instruments.


The GlycanPac AXH-1 column, based on innovative mixed-mode surface chemistry, combines both WAX and hydrophilic interaction liquid chromatography (HILIC) retention mechanisms for improved resolution of neutral and acidic glycans. The WAX functionality provides retention and selectivity for negatively charged glycans, while the HILIC mode facilitates the separation of glycans of the same charge according to their polarity and size. In addition, this column offers fast and simple charge-based separations for various glycans without discrimination of size and polarity, making it a suitable tool for  accurate quantification of glycans based on charge. The GlycanPac AXH-1 column is designed and tested for LC-fluorescence detection and LC-MS applications using volatile aqueous buffers (e.g., ammonium acetate or ammonium formate) and acetonitrile. The surface chemistry promotes retention of neutral glycans and allows elution of charged glycans at low buffer concentrations to achieve optimal MS sensitivity.
